ABSTRACT

RESUMEN El objetivo del estudio fue comparar diferentes métodos de concentración para recuperar la mayor cantidad de quistes de Giardia spp. a partir de muestras coprológicas. Se analizaron 100 muestras procedentes de hospitales de referencia nacional y se aplicaron cuatro métodos parasitológicos: concentración por sedimentación espontánea en tubo (TSET), Faust, gradiente de sucrosa de una fase y gradiente de sucrosa de dos fases. Se encontró que el método de gradiente de sucrosa de dos fases alcanzó resultados significativamente mejores en concentración de quistes (121 903 quistes/ml) y cantidad de detritos (6%), en comparación con los métodos de Faust (35 355 quistes/ml), concentración por sedimentación espontánea en tubo (20,145 quistes/ml) y gradiente de sucrosa de una fase (18 702 quistes/ml). Se concluye que el método más eficaz para la concentración y purificación de quistes de Giardia spp. a partir de muestras coprológicas es el método de gradiente de sucrosa de dos fases, lo que facilitaría los cultivos in vitro de Giardia spp.


ABSTRACT The aim of this study was to compare different methods of concentration to recover the largest number of Giardia spp. cysts from coprological samples. One hundred (100) samples from national reference hospitals were analyzed and four parasitological methods were applied: spontaneous tube sedimentation concentration (TSET), Faust, single-phase sucrose gradient, and two-phase sucrose gradient. The two-phase sucrose gradient method was found to achieve significantly better results in cyst concentration (121,903 cysts/ml) and amount of debris (6%), compared to Faust methods (35,355 cysts/ml), spontaneous tube sedimentation concentration (20,145 cysts/ml), and single-phase sucrose gradient (18,702 cysts/ml). It is concluded that the most effective method for the concentration and purification of Giardia spp. cysts from coprological samples is the two-phase sucrose gradient method, which would facilitate in vitro culture of Giardia spp.

ABSTRACT

Abstract Noroviruses (NVs) are responsible for most cases of human nonbacterial gastroenteritis worldwide. Some parameters for the purification of NV virus-like particles (VLPs) such as ease of production and yield were studied for future development of vaccines and diagnostic tools. In this study, VLPs were produced by the expression of the VP1 and VP2 gene cassette of the Brazilian NV isolate, and two purification methods were compared: cesium chloride (CsCl) gradient centrifugation and ion-exchange chromatography (IEC). IEC produced more and purer VLPs of NV compared to CsCl gradient centrifugation.

ABSTRACT

Objective. To analyze the association between daily mortality from different causes and acute exposure to particulate matter less than 10 microns in aerodynamic diameter (PM10), in Bogota, Colombia. Materials and methods. A time-series ecological study was conducted from 1998 to 2006. The association between mortality (due to different causes) and exposure was analyzed using single and distributed lag models and adjusting for potential confounders. Results. For all ages, the cumulative effect of acute mortality from all causes and respiratory causes increased 0.71% (95%CI 0.46-0.96) and 1.43% (95%CI 0.85-2.00), respectively, per 10µg/m³ increment in daily average PM10 with a lag of three days before death. Cumulative effect of mortality from cardiovascular causes was -0.03% (95%CI -0.49-0.44%) with the same lag. Conclusions. The results suggest an association between an increase in PM10 concentrations and acute mortality from all causes and respiratory causes.


Objetivo. Analizar la asociación entre la mortalidad diaria debida a distintas causas y la exposición aguda a partículas menores de 10 micras de diámetro aerodinámico (PM10), en Bogotá, Colombia. Material y métodos. Se realizó un estudio ecológico de series de tiempo (1998-2006). La asociación entre mortalidad y exposición se analizó ajustando modelos de retraso simple y retraso distribuido para diferentes causas de mortalidad. Resultados. En todas las edades, el riesgo acumulado en la mortalidad aguda por todas las causas y causa respiratoria aumentó 0.71% (IC95% 0.46-0.96) y 1.43% (IC95% 0.85-2.00), respectivamente, por incremento de 10µg/m³ en el promedio diario de PM10, tomando un retraso de tres días anteriores al deceso, mientras el riesgo acumulado en la mortalidad por causa cardiovascular fue de -0.03% (IC95% -0.49-0.44), para el mismo retraso. Conclusiones. Los resultados sugieren asociación entre el incremento de las concentraciones de PM10 y la mortalidad aguda por todas las causas y causa respiratoria.

Los mecanismos que determinan la senescencia de los eritrocitos han sido extensamente estudiados, sin embargo, no se han logrado conclusiones definitivas debido a la ausencia de una técnica que permita el aislamiento de grupos etáreos bien definidos. Los métodos más comúnmente empleados se basan en el aumento de densidad de los eritrocitos durante el envejecimiento. En este trabajo desarrollamos una técnica para la separación de glóbulos rojos de distintas edades empleando gradientes preformados de Percoll, un polímero sintético con propiedades fisicoquímicas adecuadas para trabajar con células vivas. En las suspensiones eritrocitarias obtenidas se realizaron determinaciones hematológicas, actividades de enzimas antioxidantes y el ensayo de eritrofagocitosis. Los valores de los parámetros hematológicos evaluados fueron significativamente mayores en las suspensiones de glóbulos rojos jóvenes. Las actividades enzimáticas mostraron una disminución de la capacidad antioxidante en las poblaciones de eritrocitos senescentes. Este proceso favorecería la interacción de los hematíes envejecidos con las células fagocíticas, demostrada median­te el ensayo de eritrofagocitosis. Los resultados obtenidos indican que el método de gradientes de Percoll permite una adecuada separación de las suspensiones eritrocitarias de distintas edades, con una eficiencia comparable a la observada en la técnica de centrifugación diferencial considerada de referencia.


The mechanisms that determine the senescence of the erythrocytes have been extensively studied; however. definitive conclusions have not been achieved mainly because of the lack of a technique that allows the isolation of well-defined etarian groups. The methods most commonly used for separating erythrocytes from different ages are based on the increase in density that these cells present during their aging. In the present work we have developed a technique for obtaining red blood cells from different ages using Percoll preformed gradients, a synthetic polymer with adequate physic-chemic properties to work with lives cells. In the erythrocytes suspensions we have made hematological determinations. activities of antioxidants enzymes and the essay of erythrophagocytosis. The values of the hematological parameters were significantly higher in the suspensions of young red blood cells. In the measurements of the enzymatic activity we observed a decrease of the antioxidant capacity in the populations of senescent erythrocytes. This process would promote the interaction between the old erythrocytes and the phagocyte cells, demonstrated by the erythrophagocytosis essay. The results obtained indicate that the method Percoll density gradients allows an appropriate separation of the erythrocytes suspensions of different ages with a comparable efficiency to that observed in the technique differential centrifugation, considered as reference.

ABSTRACT

Coat protein (CP) gene of sugarcane streak mosaic virus-AP isolate (SCSMV-AP) was expressed in E. coli and recombinant CP (SCSMV-AP rCP) was purified by linear sucrose density gradient centrifugation. Observation of purified SCSMV-AP rCP under electron microscope revealed the presence of potyvirus-like particles (PVLPs). The assembled particles were shown to encapsidate CP gene transcripts by slot-blot hybridization.

OBJETIVOS: Implantação de técnicas de isolamento e cultivo de células-tronco mesenquimais do sangue de cordão umbilical humano, com e sem uso de gradiente de densidade Ficoll-Paque (d=1,077g/ml). MÉTODOS: Dez amostras de sangue de cordão umbilical humano de gestação a termo foram submetidas a dois procedimentos de cultivo de células-tronco mesenquimais: sem gradiente de densidade Ficoll-Paque e com gradiente de densidade. As células foram semeadas em frascos de 25cm² a uma densidade de 1x10(7)células nucleadas/cm² (sem Ficoll) e 1,0x10(6) células mononucleares/cm² (com Ficoll). As células aderentes foram submetidas a marcação citoquímica com fosfatase ácida e reativo de Schiff. RESULTADOS: No procedimento sem gradiente de densidade Ficoll, foram obtidas 2,0-13,0x10(7) células nucleadas (mediana=2,35x10(7)) e, no procedimento com gradiente de densidade Ficoll, foram obtidas 3,7-15,7x10(6) células mononucleares (mediana=7,2x10(6)). Em todas as culturas foram observadas células aderentes 24 horas após o início de cultivo. As células apresentaram morfologias fibroblastóides ou epitelióides. Na maioria das culturas houve proliferação celular nas primeiras semanas de cultivo, mas após a segunda semana, somente três culturas provenientes do método sem gradiente de densidade Ficoll-Paque mantiveram crescimento celular, formando focos confluentes de células. Essas culturas foram submetidas a várias etapas de tripsinização para espalhamento ou subdivisão e permaneceram em cultivo por períodos que variaram de dois a três meses. CONCLUSÃO: Nas amostras estudadas, o isolamento e cultivo de células-tronco mesenquimais do sangue de cordão umbilical humano pelo método sem gradiente de densidade Ficoll-Paque foi mais eficiente do que o método com gradiente de densidade Ficoll-Paque.


OBJECTIVES: Implantation of cell separation and mesenchymal stem cell culture techniques from human umbilical cord blood with and without using the Ficoll-Paque gradient density method (d=1.077g/ml). METHODS: Ten samples of the umbilical cord blood obtained from full-term deliveries were submitted to two different procedures of mesenchymal stem cell culture: a) Method without the Ficoll-Paque density gradient, which concentrates all nucleated cells; b) Method with the Ficoll-Paque density gradient, which selects only low-density mononuclear cells. Cells were initially plated into 25 cm² cultures flasks at a density of 1x10(7) nucleated cells/cm² and 1x10(6) mononuclear cells/cm². RESULTS: It was obtained 2-13x10(7) (median = 2.35x10(7)) nucleated cells/cm² by the method without the Ficoll-Paque gradient density, and 3.7-15.7x10(6) (median = 7.2x10(6)) mononuclear cells/cm² by the method with the Ficoll-Paque gradient density. In all cultures adherent cells were observed 24 hours after being cultured. Cells presented fibroblastoid and epithelioid morphology. In most of the cultures, cell proliferation occurred in the first week, but after the second week only some cultures - derived from the method without the Ficoll-Paque gradient density - maintained the growth rate reaching confluence. Those cultures were submitted to trypsinization with 0.25 percent trypsin/EDTA solution and cultured for two to three months. CONCLUSION: In the samples analyzed, cell separation and mesenchymal stem cell culture techniques from human umbilical cord blood by the method without the Ficoll-Paque density gradient was more efficient than the method with the Ficoll-Paque density gradient.

No presente trabalho, foi avaliada a influência da concentração e de diferentes conbinações de tratamentos do hidrolisado hemicelulósico ácido de bagaço de cana-de-açúcar, sobre atividade da enzima xilose redutase (XR) de Candida guilliermondii. Em paralelo, determinou-se a atividade da enzima xilitol deshidrogenase (XD) presente nas células, já que o nível de atividade desta enzima pode interferir no rendimento em xilitol, quando a bioconversão é feita por via fermentativa. As células foram cultivadas nos hidrolisados provinientes das diferentes combinaçõoes de tratamentos pela alteração do pH utilizando bases e ácidos e adsorção com carvão ativo. ABSTRACT

We describe a simple method using percoll gradient for isolation of highly enriched human monocytes. High numbers of fully functional cells are obtained from whole blood or buffy coat cells. The use of simple laboratory equipment and a relatively cheap reagent makes the described method a convenient approach to obtaining human monocytes.

ABSTRACT

A Ficoll-Hypaque gradient centrifugation technique was used for isolation and concentration of microfilaria from peripheral blood of 30 subjects with clinically and parasitologically diagnosed Wuchereria bancrofti infections. 86% of the microfilaria were found in the Ficoll-Hypaque layer. None were detected in the plasma, leucocyte layer or lower erythrocyte layer. 14% of microfilaria were identified on the top part of the erythrocyte layer. A 35-fold concentration and 88% quantitative recovery of parasites was achieved by conventional centrifugation of microfilaria-rich Ficoll-Hypaque layer. Following the centrifugation procedures, living motile microfilaria were separated. These results indicated that Ficoll- Hypaque centrifugation technique could be an effective method for the detection of low levels of microfilaremia, and for obtaining relatively pure suspensions of living microfilaria for metabolic studies, production of antigen-rich excretory-secretory products and antigen analysis

ABSTRACT

Phospholipid composition of sheep blood platelets and its various plasma membrane fractions have been analyzed. Based on their flotation rates in discontinuous sucrose density gradient centrifugation, three membrane fractions were isolated. 5'-Nucelotidase and alkaline phosphatase were distributed nearly equally in all the three membrane fractions. However these membrane fractions showed differences in the distribution of phosphatidyl ethanolamine, phosphatidyl choline and phosphoinositides. Phosphatidyl ethanolamine was predominant in fraction I (11.05 micrograms PLP/mg protein) while phosphatidyl choline was predominant in fractions II and III (110.10 and 68.30 micrograms PLP/mg protein respectively). Phosphatidyl inositol (Ptd-InsP) was equally distributed in all three membrane fractions. However, both Ptd-InsP and phosphatidyl inositol 4,5-bisphosphate were about 4-fold higher in fraction II (73.55 and 89.89 micrograms PLP/mg protein respectively).
